Laboratory Techniques

Hello I've just started doing a bioscience course in which we have a graded unit where we are free to choose our own project. As keen fermenter I was hoping to analyse some miso's. My plan is to make around four miso's all the same but with different salt levels. I am hoping to look at things like enzyme activity, glutamate levels, protein levels and lactic acid levels.

My problem is I have zero experience in a lab (15 years in kitchens though haha) thanks to COVID stopping us going in the labs. I was wondering if any one here could help direct me in what tests, protocols or techniques I could use?

Any help would be greatly appreciated as I want to use to chance to examine miso but don't really know where to start

Textbook on alloying laboratory techniques

I'm doing Masters degree in Material Science studying the segregation and diffusion in Cu-Ag alloys. Normally we would evaporate silver onto copper samples and then anneal the samples for several days to obtain homogeneous alloys. My professor wants me to cast a small billet of my alloy instead. I want to read up on some alloying techniques. Are there any textbooks or papers you can recommend? I need a high purity, high homogeneity product.

Has anyone done Science & Laboratory Techniques (Level 5) at St. John's?

Let me explain that I have not done any type of science study since 2nd year in school. I'm almost 30 now and consider my scientific knowledge very limited. I am however interested in learning and think working in a lab could be very interesting and engaging. I particularly have an interest in biology. My last job was in software development (i have a level 6 cert in software) but i don't particularly enjoy that work.

I'm looking for help/advice from someone more experienced in these fields and perhaps anyone who has done the above mentioned course.

Is it difficult to get into? Is it ok to start studying something like this at level 5 without much prior knowledge? What is work in a lab environment like?

How do I increase my speed in laboratory techniques?

I have recently been hired as a scientific officer at a pathology. I've been at the laboratory for approximately one month, however I'm struggling with picking up speed when it comes to processing samples.

Techniques that are used include plate streaking, and culturing/pouring urines into tubes for analysis. I have definitely improved since I first started, although it feels like I've hit a plateau.

Does anyone have any suggestions to increase speed in techniques such as these?

Comparison of Antioxidant Activity and Extraction Techniques for Commercially and Laboratory Prepared Extracts from Six Mushroom Species , study [ 03 - 2021 ]

Highlights

I. obliquus and G. frondosa outperformed other mushrooms in five antioxidant assays.

Laboratory-prepared mushroom extracts exhibited higher AOX activity than commercial extracts.

Preliminary stability tests on laboratory-prepared chaga demonstrated a stable AOX activity after 1-month, and > 80% activity loss after 6 months.

AOX activity monitoring during a 1-month commercial extraction process showed no increase after 1 day.

Our data have important implications for consumers and the natural product industry.

Abstract

Mushrooms are a potent source of dietary antioxidants and are often marketed for their capability of decreasing oxidative stress and preventing disease.

To date, however, little research has been done on the antioxidant activity of commercially available mushroom extractions, or their stability over time.

Herein, the antioxidant capacity of six commercially available mushroom tinctures (hydro-alcoholic extracts of chaga, maitake, shiitake, reishi, lion’s mane, and turkey tail) were evaluated using a panel of five antioxidant assays: Oxygen Radical Absorbance Capacity (ORAC), Nanoceria Reducing Antioxidant Capacity (NanoCerac), DPPH radical scavenging, Total Phenolic Content (TPC), and Ferric Reducing Antioxidant Power (FRAP).

Commercial samples were compared to our own laboratory preparations of the same species, in a preliminary assessment of extraction technique and antioxidant stability.

Mushroom extracts exhibited different antioxidant capacities depending on species and extraction method (e.g., 0.023 to 0.63 umol TE/mg for commercial ORAC vs. 0.05 to 1.8 umol TE/mg for laboratory ORAC for all species). In all cases, laboratory extracts exhibited considerably higher antioxidant activities than commercial extracts.

In both extract types, chaga and maitake consistently demonstrated higher antioxidant activity than the other mushrooms. Preliminary antioxidant stability testing of our samples revealed stable ORAC values for one month at room temperature, followed by a 48% decrease after 4 months.

Conclusions: Our results provide important insights regarding the effects of extraction techniques and storage conditions, on the antioxidant activity in mushroom extracts, particularly those made and sold commercially.

I just came across a question that required you to know not only what Southern Blot is used for (DNA Analysis), but it required in depth knowledge of the laboratory procedural steps involved in doing the technique.
