tl;dr - The Hunter's Rune was probably a literal depiction of a Hanged Man to Pthumerians. They may have originally used them to drain the heaviest parts of blood, its impurities, from dead/stagnant bodies. Those impurities were Blood Echoes which is what gave them superhuman powers elevating them beyond humanity.
There have been many threads where others have pointed out the similarities between the Hunter's Rune and the Hanged Man tarot card. I don't intend to go too deeply onto the subject of tarot specifically, but rather focus on contextualizing some Japanese cultural notes and how they may be applied to Bloodborne.
Note: Much of this is based on conjecture upon possible interpretations of the game's more open-ended mechanics/story.
In many ways, Bloodborne draws on many of the same themes used within Sekiro--of how stagnation can become a source of strength and corruption. Sekiro is obviously much more overt with this theme, but it's also present in Bloodborne, though perhaps obscured a bit by localization and general abstraction.
To clarify some of what I mean by that, I'd argue the terms used for Blood Dregs, the Corruption rune, and the Impurity rune are different from what's presented in the Japanese version of the game.
Cainhurst's "Corruption" rune comes from ็ฉขใ kegare. Kegare is hard to translate directly into English, but I would have opted to call it "Impurity." ็ฉขใ kegare is linked to concepts of dirtiness, filthiness, unholiness, defilement, and corruption, though it's not limited to a physical nature. It can be transcendent with certain acts able to make someone kegare. This concept also plays into the imagery of filthy things in Japanese culture, like bugs and other creepy crawlers. This is partly why things like centipedes as Vermin in Bloodborne or parasites within Sekiro are so prevalent as a theme within FROM games.
Blood Dregs as I mentioned before comes from ่กใฎ็ฉขใ chi no kegare, which means the "kegare (aka impurity) of blood." This change in meaning isn't too terrible, but it is worth noting how much more direct the connections in Japanese are between Blood Dregs and the Cainhurst "Corruption" rune.
One of the bigger changes in intended meanings comes from the League's "Impurity" rune. In Japanese, it's ๆทใฟ yodomi, which according to jisho.org means, "stagnation; deposit; sediment; backwaterโ." I would personally translate it as something clos

I keep a Tarot deck on my headboard for whenever. Right when I got home I found one of the cards laying on my pillow, scared the living shit out of me.
