Hematoxylin Puns

I am tired. No matter how many pencils i buy, it keeeeeps on breaking. Its getting on my nerves. Does anybody know a good brand E&H that doesn't break easily? I think I've spent more on these pencils than food this month. Please help

So, basically, there's a brand new patomorphology department in our lab and I was thrown there like a blind kitten. I've read a few books and some stuff online, but still can't find the right staining protocol for our samples. Nuclei don't get stained at all, and I tried to keep them in hematoxylin for longer periods of time, but still. Would someone be willing to share theirs for me to try, ot any general tips and tricks?

I'm willing to take some courses obviously, when I find any available (maybe there are some online ones you could recommend?) but I'm required to provide immediate results, unfortunately.

I know that the best advice would be leaving the place, but it's not an option:'D

I'm trying to optimize my staining technique for a HE stain to count epithelial cells and measure crypt height for colon tissues.

I currently have Mayer's in my lab and would like to keep using it to avoid purchasing more reagents but the blue colour seems to wash out after adding eosin.

I was recommended the Richard Allan Scientific 7211, does anyone have experience with this?

Since I'm using an alcoholic eosin, I think I might need to include an alcohol rinse step before counterstaining.

But I thought I'd ask around before spending more time on this since I'm in a bit of a time crunch :(

So I have been doing immuno on some paraffin-embedded sections and my hematoxylin nuclear counterstain comes out very light. I see publications with bright blue staining and mine is more or less light blue/lavender in color.

Here is my current protocol. After color development with DAB-Plus kit (Invitrogen):

1. 8 minutes Harris modified hematoxylin (Sigma)

2. 5 min running tap

3. 30 sec 2% acid alcohol (this is 70% EtOH, 29% water, 1% HCl)

4. 1 min running tap

5. 60 sec 0.2% ammonia (200ul ammonium hydroxide in 100 ml water)

6. 5 min running tap, then dehydrate.

Would love any and all suggestions!

Does anyone have any preferance over either option? My lab is currently working with ammonia, but Im trying to find some testimony as to which is preferred.

im trying to stain xenograft tumor sections (3-4 um) for collagen using Gomori's trichrome stain procedure. The collagen stain is working well, but the hematoxylin (Wiegert's iron) is washing off after i put the slides in acetic acid for differentiating the stain. I make the working solution fresh every time, so not sure what may be going on. Any suggestions would be greatly appreciated!

We are doing mitosis in my high school class, and currently I have the students counting cells in various stages in a root tip. The slides we use are ones I ordered from Flinn about two years ago. Some of them are OK, but a lot are really crappy and hard to see. I want to order some better ones. I am considering ordering from Carolina, but I'm wondering if I should order the slides with the hematoxylin (same as what I have now) or the quad stain. Has anybody ordered these or other slides and had good/bad results?

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.

Again, to all the fellow dads, I apologise if I'm sounding too harsh. But I just needed to get it off my chest.

For context I'm a Refuse Driver (Garbage man) & today I was on food waste. After I'd tipped I was checking the wagon for any defects when I spotted a lone pea balanced on the lifts.

I said "hey look, an escaPEA"

No one near me but it didn't half make me laugh for a good hour or so!
