I've done 40/100 pulls. the shops still has 46(aupply of 250 HC) available and another 4 pulls left from clearing babel tower and black flash for the first time which i have yet to claimed. So that's a total of (40+46+4) 90pulls available. Ive checked all the avaiable unclaimed rewards and i cant seem to find where i get the HC for the last 10 pulls i need for guarantee.

Or am i missing something?. Any info would be nice.

Hey guys I'm new to the community and my sexuality became evident for me just yesterday. Everything seems to make sense now except for my preferences and my relationship with sexual culture.

You see when I was a preteen I saw porn for the first time and even though I enjoy it I didn't masturbated until I was 18 for the first time. I never had a desire to do so until I was in an active relationship, but I seems once I am with someone my Libido skyrockets wtih the person I'm dating.

The confusing parts come with the following:

With the years things changed, I still enjoy porn and do masturbate from time to time, but I don't feel arose if a male or penetration are/is involved in set porn, (and before you ask, lesbian porn doesn't interest me either) although I am only romanticaly interested in men.

Another confusing part is that sex, being sexy and the ongoing sexy culture are topics that I find fascinating and love to engage in, as a matter a fact everybody saw me as this sexy girl that is super active since my personality can come off as hypersexual. I assure you, despite all this is impossible for me to have sexual desire for someone that I'm not in love with, In fact, I will dare to say I start being open to the idea of sex after I am in love instead of being sexually attracted until much later in the romantic relationship, but due to my personality I just thought I was just a very picky romantic, and taking it slow type of heterosexual.

Lastly I do have a type, I feel attraction to men that look a certain way, only men, before I was open to the the idea of girls but didn't find I could be romantic with a lady, and all the definitions I see talk about gender not being important for demis, is this true or is there such thing as a heteromantic demisexual?

I don't know if this is normal, all the experiences I see here come from people that don't see gender and don't see looks and don't form part of the sexual culture. Can someone help me make sense of this?

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.

Again, to all the fellow dads, I apologise if I'm sounding too harsh. But I just needed to get it off my chest.

This is yet another short chapter, but it does provide us with some interesting insights into the Punishing Virus. So let’s go.

Story Recap

We learned in Frozen Darkness that the International Space Station (ISS) was infected by the Punishing Virus. The infection occurred soon after the virus outbreak on Earth. The virus was sent into space during the explosion of the Zero Point Energy reactor. ISS and Babylonia had been having constant conflicts, which was also indicated in the Frozen Darkness. So at the beginning of this chapter, Babylonia decided to launch an all-out attack and end this threat once and for all.

In order to better understand the story, we actually need to start from Watanabe Astral’s Interlude. The time of this interlude was during the Great Retreat (aka Operation Acadia), we learned that Watanabe was a pilot of Babylonia at that time. In one mission, he was working with an legendary flying-ace Cyril. Cyril is the father of Bruce, who was Watanabe’s former teammate who were killed by the Corrupted during a rescue mission (this story is in Watanabe’s first interlude). In this mission, they needed to escort a collection of “essential supplies” for Babylonia, according to President Hassen. However, during the mission, Cyril betrayed Babylonia and started attacking Watanabe and the transport fleet. Watanabe fought back and eventually defeated Cyril. But before Cyril died, he told Watanabe the truth about those “essential supplies” that they tried to protect: they are nothing but a bunch of luxury items for the rich and powerful that live in Babylonia. He couldn’t accept that they are actually wasting time, resources, critical space on Babylonia for things like this. Therefore, he doesn’t want to fight for this corrupted World Government any longer, and that’s why he betrayed them.

After defeating Cyril, Watanabe met with President Hassen. Clearly Watanabe’s loyalty to the World Government was severely shaken by Cyril’s words, so he asked to leave Babylonia and be sent back to Earth. Hassen reluctantly agreed, and he knew this action would never have been approved by the World Government (because Constructs are elite military forces), so he kept this off the record. Watanabe changed back into his old Nightblade frame and snuck into a transport dumping damaged Construct frames on Earth, where he eventually formed the Forsaken. Since this agreement was kept off the book, Watanabe was marked defected by the Purifying Force and being hunte

For context I'm a Refuse Driver (Garbage man) & today I was on food waste. After I'd tipped I was checking the wagon for any defects when I spotted a lone pea balanced on the lifts.

As you all may know, the roseTTA fold algorithm published over the summer can predict heterometic protein complexes. I am looking to predict the structure of an unsolved heteromer for some study within my lab.

In the science article, they use it to predict theoretical structures of heterotrimers (https://www.science.org/doi/10.1126/science.abj8754) , however upon looking in their github (https://github.com/RosettaCommons/RoseTTAFold) they only seem to have prodcedure for predicting complexes of heterodimers. I've successfully done the procedure to predict heterodimers of proteins of interest to our lab, but cannot figure out how to predict trimers from the github

Does anyone know the procedure for predicting trimers or where I can find it?

if this isn't the best sub for this please point me to the right place.

Testing next week and lowkey freaking out that I've been thinking about this wrong. I always thought that since SDS denatures the protein, it separates them based on size, but since it is non reducing, it wouldn't separate any dimers/heteromers etc because I thought SDS doesn't break disulfide bonds that are responsible for the attachment of two subunits together, which is why I chose C because since the question says non reducing, I thought no info can be deduced about covalent linkage from it. I do understand why D is correct, but not why C is incorrect.

Can someone please specify the difference between reducing vs non reducing gel in the context of this question, like what would be different in these gels if we had used the reducing gel instead? Thank you so much!!

Protein X is a tetramer composed of 2 identical heterodimers because while in gel B (native gel) Protein X runs as a single band of approximately 140 kDa, in gel A (non-reducing SDS-PAGE) it is denatured into 2 large subunits of 50 kDa each and 2 small subunits of 20 kDa each. The two subunits of the heterodimers in the variant form are covalently linked because gel A (non-reducing SDS-PAGE) shows the two subunits run as a single band at approximately 70 kDa.
