On Thursday, I will have completed a two-week treatment of kanamycin/kanaplex for my betta fish. She is not getting worse, but she has not consistently improved. She still has a great appetite and a lot of energy. But she has pineconing on her whole body and dull coloring.
I just started doing Epsom salt baths yesterday and I'm going to try to do that 1-2x a day. I also have aquarium salt in the quarantine tank at about 1/2tsp per gallon. I've been changing about 25% of the water daily, give or take.
Is there anything else I can treat her with after kanamycin? I am worried about doing another antibiotic; I don't want to create antibiotic resistant bacteria.

Hey guys, I was wondering if it is possible to inoculate LB media and store the samples at 4 degrees for 24 hours, then grow them overnight to be used the following day? Reason being I can't get into the lab tomorrow, but I don't want to lose a day of work on Friday due to a lack of grown culture. I'm growing these cultures to miniprep, and have someone in the lab who is willing to move my cultures to the shaker from the fridge tomorrow. Any suggestions would be greatly appreciated!
I have a problem with my left eye and the doctor prescribed me Kanamycin Sulfate H (that's what is writen on the box). It's kind of like a cream that I need to put in my eye. I also take 2 others types of eye drops but this is the only antibiotic. Can I drink alcohol?
Just an FYI for anyone that is in the horrible spot I was in the past month. I had a pretty bad columnaris outbreak in a 55g tank that houses mostly guppies but also a healthy community of crystal reds. I bought the CRS on reddit about 6 months ago as juvies and they were just beginning to take off. Breeding etc.
Well- the columnaris was doing an absolute job on my guppie population (neons as well). I lost about 70% of the fish I had. I knew it had to be treated but all of the treatments call for Furan-2 and Kanamycin. I couldn't find ANY solid info on Furan-2. Some posts said it would kill shrimp etc.
Just got done with essentially a week of heavy treatment and the CRS are doing absolutely fine. Didn't lose one. Babies are all good. Guppies are healthy again. Everyone is happy.
Furan-2 was safe for me.
20ug/mL gave me growth from untransformed dh5a. 50 ug/mL didn't give me any transformed colonies. Tips for transforming on kanamycin plates?
Hey folks, I've been having some trouble with the mutagenesis process using a kanamycin-resistant vector. In my amp-containing vectors, the mutagenesis, transformation of mutagenic PCR products, and miniprepping of that transformation all works great but when I use my Kan vector I get very few or small transformant colonies and poor, impure miniprep yield. I've been using 50 ug/mL kan, and I just started one using 25, so we'll see if that helps. Have any of you had a similar problem, and how were you able to fix it?
We're having trouble getting our plasmid after a transformation and think that LB Agar kanamycin plates made in November of last year may be the problem.

Basically what I'm seeing is that the dicotyledons are dark green (instead of yellow like they are being fully susceptible), but the first pair of true leaves is yellowish and doesn't look healthy.
Fully resistant plants should be dark green on all leaves. You can see a better example of what I'm talking about when I zoom in - there are a few totally resistant plants in there, and then a bunch with the weird half-resistance.
Kanamycin resistance isn't dose dependent so this isn't a case of some of the plants being heterozygous and some being homozygous.
This is a T2 generation, which means 75% of the plants should be resistant to kanamycin. As you can see from the first image, most of the plants have this weird paleness on the second pair of leaves, some (<25%) look like fully resistant and a few (one or two) are fully susceptible.
I have 5 plates out of 10 that look like this, one plate that had the proper 75-25 proportions of living:dead, but the living plants still mostly had the yellowness on the true leaves. The remaining plates mostly didn't germinate; the seeds that did were fully resistant. (I think that's a problem with my seeds, not the plates).
Is it a nutrient deficiency in my agar medium? We're using MS salts, sucrose, and agar in the recommended proportions. Our salts are ~ 2 years old and have been stored in a dessication box in the fridge.

I'm pretty sure that two of my bettas have the slower growing variety of columnaris. The problem is trying to figure out how to treat it. Seachem wants me to dose the food, but one is a very poor eater and I'm not certain this is going to work.
Any suggestions on dosing with Furan and Kanamycin. I'm seeing so much information I'm having trouble making sense of it all.
