Hello, I'm wondering if anyone knows how pen/strep would affect collagenase type 1 activity? I am doing a collagenase activity assay on a collagenase solution with pen/strep.
Would it be possible to tenderize meat using some enzyme?
As i understand it collagen is what makes meat tough so could you use an enzyme to break it down to some degree, and is this why people use pinapples to tenderize meat?
Hi Labrats! I need to reconstitute collagenase IA lyophilized powder to prepare a digestion buffer for isolating tumor cells from breast samples. The manufacturer (Sigma) advises to use Krebs Ringer buffer with calcium and BSA or in Hank's Balanced Salts for tissue culture. However I've seen a protocol that uses PBS instead. Would you recommend doing so? Also, do you follow any special safety measures besides wearing gloves and working in a laminar flow hood?
I also have to reconstitute hyaluronidase for the same purpose. The product information sheet advises 20 mM sodium phosphate buffer, pH 7.0, with 77 mM sodium chloride and 0.1 mg/ml BSA at 1 mg/ml. Would it be the same to use just PBS?

Hi labrats, I'm looking to use collagenase D for my experiment. I reconstitute it in Hank's (minus calcium and magnesium), and the TDS from Sigma-Aldrich says you can only store it in this form up to a week at -15 to -25 degrees Celsius before it loses most of its activity (PDF link: http://www.sigmaaldrich.com/content/dam/sigma-aldrich/docs/Roche/Bulletin/1/colldrobul.pdf).
The enzyme is shipped in a dry powder at 100 mg. The problem is, it's troublesome and imprecise to reconstitute it unless I weigh out at least 25 mg or so to use, and I don't want to have to reorder the enzyme every month or so (it's shared with other lab members). Any suggestions?

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.
Again, to all the fellow dads, I apologise if I'm sounding too harsh. But I just needed to get it off my chest.

For context I'm a Refuse Driver (Garbage man) & today I was on food waste. After I'd tipped I was checking the wagon for any defects when I spotted a lone pea balanced on the lifts.
