Adenosine‐to‐inosine (A‐to‐I) editing is a conserved RNA modification that contributes to immune response and overall cellular function. RNA editing patterns can vary significantly between different cell and tissue types, and hyperactive A‐to‐I signatures are indicative of several diseases, including cancer and autoimmune disorders. Because of the importance of these differences, there is significant need for efficient methods to measure overall A‐to‐I editing levels in cellular RNA. The current standard approach relies on RNA‐seq to indirectly detect editing sites, which requires large investments in time and material as well as extensive computational analysis. Here, we utilize Endonuclease V (EndoV), which binds to inosine in RNA, to develop a protein‐based chemiluminescent bioassay to directly profile A‐to‐I editing activity. We previously showed that EndoV can enrich edited transcripts prior to RNA‐seq, and we now leverage this activity to construct an EndoV‐linked immunosorbency assay (EndoVLISA) as a rapid chemiluminescent method for measuring global A‐to‐I editing signatures in cellular RNA. We first optimize and validate our assay, illustrating highly selective and sensitive detection of inosine in RNA. We then demonstrate rapid detection of inosine content in treated cell lines, demonstrating equivalent performance against current standard RNA‐seq approaches. Lastly, we deploy our EndoVLISA for profiling differential A‐to‐I RNA editing signatures in normal and diseased human tissue, illustrating the utility of our platform as a diagnostic bioassay. Together, the EndoVLISA method is cost‐effective, straightforward, and utilizes common laboratory equipment, offering a highly accessible new approach for studying A‐to‐I editing. Moreover, the multi‐well plate format makes this the first assay amenable for direct high‐throughput quantification of A‐to‐I editing for applications in disease detection and drug development.

I know of molecular cloning - a lab manual. However, I want something that is more bioanalytical, not just obtaining replicates of genetic info as much. More looking for bioassay protocols for the general cell activities listed above.

I have an organometallic compound that I want to perform a bioassay on, procedure involves dissolving compound in pure DMSO first before diluting it down with buffer to obtain a solution used for treatment. However upon dilution the compound precipitates out as fine particles which screws up the assay results - any idea on how to solve this without changing the ratio of DMSO to water? Also in the assay this solution is added to more water (buffer), which causes even more precipitation.
Helpful info: Compound solubility is approximately 0.001g/mL in DMSO, practically insoluble in water

There’s something fascinating about doing normal things on psychedelics. From movie theaters to airplanes, I have incorporated low doses of LSD into daily life (though far from on a daily basis) for over a year now, however physical activity was something I had never tried while tripping, micro-dose or otherwise. The idea came to me one night - late - as I was cleaning my apartment on a +-50ug dose (all “doses” are estimated by dividing up 150ug tabs and as always should be considered merely as approximations). For several hours I cleaned and organized while my significant other slept slept peacefully in the bedroom of our converted warehouse loft.
I was in a fantastic mood - full of that magical, never-ending energy you only have on those really rare, perfect days - and I remember thinking to myself “I feel as though I could run a marathon right now”. From that moment on, running a 5K on LSD was on my mind and I would only have to wait 3 months for my opportunity. The following is what I wrote up the evening after the race.
I wake up feeling flawless. An enjoyable night spent in deep conversation with a good friend the evening before had ensured a restful night of sleep, despite the absence of my partner - out of town on business - something that usually throws both of our sleep patterns. I had prepared everything I needed the night before - a third of a (claimed) 150ug tab of LSD, a Xanax in case of sincerely needing to abort (though I feared little chance of that happening at this dosage), and finally a Dramamine to counter the initial nausea I sometimes experience taking Lucy.

As this will be my first public bioassay I feel as though some background might be warranted. I have experimented with various drugs, particularly those of a psychedelic nature, for over 15 years now. I had always been warned about the dangers of using drugs and how they would destroy my future, and yet, here I was integrating them into my life both as a means of self-exploration but also for genuine scientific inquiry. As a sufferer of chronic migraines I have followed the evolution of psychedelic migraine treatments with great interest for some time and not willing to wait for policy to catch up with science, I have successfully self-administered LSD and Psilocybin for unprecedented pain relief. Not only was I finding that psychedelics actually improved my health, but I was successful in my career in Marketing, eyeing a potential return to university for a PhD in Psychology and generally the healthiest and happiest I had ever been. I wasn’t taking these compounds as a means of escape, they didn’t lead to self-destructive behaviors, nor was I suffering from addiction - my eyes had been opened to the fact that the war on drugs, particularly the physically harmless ones like LSD and Psilocybin, was political theater - a thinly veiled attempt at control.
Many might question my motives - I have been asked to provide a moral rationalization of my experimentations on several occasions. In my defense, not that it is needed, I can only say that I have a passionately inquisitive scientific mind and an immovable belief in individual freedom. I do not believe the government should have the right to dictate our consumption, particularly the consumption of plants, fungi and drugs that have a positive effect on people and in some cases make their lives more liveable (as a chronic migraine suffered I fall in that category). In any case, I was working on seeing the physical world (32 countries now) but just as profoundly exciting, I believe, is the ability for man to make an inner journey. ... keep reading on reddit ➡The following is a report concerning an attempt of mine to produce a Tryptamine Alkaloid + MAOI brew out of Phalaris grass and Syrian Rue. I'm recording my thoughts towards this experiment as well as the process used and a trip report of the brew itself. Hopefully some of you might find the results of my reckless grass tea guzzling entertaining and perhaps even informative.
First, allow me to preface with some info about this unlikely source of psychedelic material, and what inspired me to attempt this whole thing. As a hallucinogen enthusiast and armchair ethnobotanist, I was delighted to discover recently that a genus of grasses known as 'Phalaris' have been found to contain psychedelic tryptamines. I was even more delighted to discover that these grasses were native to my area, and a quick stroll along the riverside by my home provided some samples which confirmed this.
As you shall soon see, this little experiment was a somewhat risky endeavor, but before you flood my inbox with (admittedly deserved) messages calling me an idiot, allow me to at least say that I tried to do my homework on this one. Upon realizing there was a psychedelic plant I had unknowingly walked past hundreds of times, I began to scour the internet for more information. A number of threads on the DMT Nexus provided accounts of several enterprising psychonaut's attempts to perform A/B extractions of Phalaris, with mixed but overall positive results. I also came across several Erowid experience reports detailing user's consumption episodes, ranging from extractions, brews, or smoking the grass raw. Some reported little to no effects, while others indicated that the grass was capable of inducing quite strong experiences. The most interesting document I came across was a write-up from 1994 about the alkaloid contents of Phalaris, along with a subsequent rue based ayahuasca bioassy. Having compiled enough information to prove that at least some people had gotten off with this stuff, I dutifully grabbed a knife and set out to chop some grass.
