I took a few genetics courses in college but my knowledge on the subject is still pretty basic. Nonetheless, immortal cell lines have fascinated me recently.
It could be that his name would be implemented as a new word in the dwarf language (storyteller/raconteur/chronicler etc). Or, perhaps one of his intro soundtracks could be included in some part of the music for DF. Whatever form the tribute might take, Krugg has really given so much and worked so hard for the community, and his content is available freely on YT, despite its exquisite quality. Heβs brought DF to life for so many new and old fans alike. I think he deserves it. Iβd love to hear what you think of this idea, and how it might be done.

I'm dropping from 4 lines to 2. I have T-Mobile on an old plan that was a great deal but now I'm thinking of trying Mintmobile, since they use T-Mobile's service but are cheaper.
Checking our usage, we are never over .5 GB because of Covid I guess (always at home).
Has anybody switched to Mint and have you had any problems? How is traveling to Europe?
UPDATE: Mint is having a buy 3 months get 3 months free special so I went for it. Hopefully it will work out because it's so much cheaper!

Highest profit %. Those diamond handed mother fuckers that held those shares from eons ago, posting a 1,000,000,000% gain on a single sale. This is anyone, from x to xxxxx that have had the bollocks to hold that single buyin that is now single handily gonna fire them to the moon.
I can see it now, new high score guy posting arcade screens of the new daily winner, all whilst the float is locked and people are selling their lives (shares) on brokers to achieve the ultimate post porn after it is all said and done.
But then, out of the shadows, after the title screen has rolled, a secret achievement and high score is revealed.
The morale of the story is once this kicks off itβs going to be fun, it needs to stay fun and the aim of the game is to high score.

So i've been paying well over $80/month for my Verizon line from years ago. It came with unlimited data, talk, text, as well as perks, but I just came across a few other companies that offer a lower tier such as Mint $30 for unlimited everything or Visible and realized that although they use the same antennas on Verizon their speeds are extremely slow! I'm a Uber driver and the speed is just ridiculous where it won't even load google maps. Fast forward a few weeks - my friend told me about Circledin. I was able to keep my same Verizon line but instead of paying $80, I now pay $40/month by joining a family plan! I'm saving over $400 and wanted to share this awesome service.
You might want to check out Circledin and carriers such as Mint or Visible. I wish i knew about this before paying an arm and a leg for my cell phone bill. I hope this helps the Frugal community!
I'd like to hear experienced home growers share their thoughts about what Paul says in his book Growing Gourmet and Medicinal Mushrooms, pages 97-100.
He basically says that even fungi need to sexually reproduce (make spores, right?) every once in a while to prevent cell lines from dying out. Meaning if you only clone and/or transfer from medium to medium, the further you get from the genetic origins, the weaker the strain becomes and eventually doesn't perform that well anymore.
He says that he doesn't go over 10 cycles of transfers/cloning and works with culture slats to stay as close as possible to the genetic origin of that strain. There needs to be some cycles, obviously, to isolate the good traits from the bad.
I'm interested in hearing any thoughts on this in a homegrow environment, since Paul is coming from a commercial point of view. Specifically cubensis growers, but any other species interests me as well.
Do you agree with the max 10 cycles line? Have I understood the subject correctly? How do you personally approach the challenge of staying close to the genetic origin, while still isolating the best traits of your strain? Any stories or anecdotes are welcome π Thanks
Title says it all. My team received a sample of precious cell lines from another site, and we saw a large loss after thaw. We handle a lot of frozen cell lines, and this was the first one that totally crashed and burned on us. We hopped on a call with the other site to start troubleshooting how to minimize loss. After outlining a cryo-recovery method development plan, we decided that the method of last resort would be to ship the cell lines live and with heating pads.
The study director reminisced about the good 'ole days 22 years ago before 9/11. Her staff would walk onto trans Atlantic flights with fragile cell cultures strapped to their bodies to keep a consistent warm temperature. What happens if security gets suspicious? Well, you were supposed to claim it was a health tonic and happily drink one of the flasks, sacrificing one of your tubes in order to save the rest. They did a small test in advance and determined that drinking ~50 mL of stem cell culture media does not harm the average person in any way. Ahh, those were the days.

I'm trying to find out if our human cell line expresses a protein (and the levels, if available) before transfection and I'm not sure how to go about looking for that information. Wondering if any of you have ideas.
I know about CCLE but that's for gene expression and also somehow didn't find my protein (gene) of interest there for our cell line.
