Self-Experiments with Sublingual Administration of Liquified Mitragynine Concentrate (80%, 100mg/1mL)

So for a while I have been curious about the possibility of sublingual administration.

The short version was I believe that it is possible, with rapid uptake (<15 minutes) but shorter action versus oral consumption of the same sample.

I obtained a preparation of 80% mitragynine (0.8% 7-OH-MG), equating to 100mg mitragynine per 1mL of solution. My normal dose (recently) has been ~93mg per dose (1.86% x 5 grams) of raw plant material. This preparation had slightly more 7-OH-MG than what I am used to, but without artificial supplementation, and less of the other minor alkaloids.

Test 1

I stopped all kratom use until I had baseline pain, and was beginning to experience withdrawal, at about 12 hours (which is normal for my experience given that I use multiple times a day for chronic pain). I took all my normal medications accept for a muscle relaxant which I sometimes use.

On the first day, I drew 1mL of solution (+/-0.1mL) and put directly into the back of my mouth, swallowed, and then swished it around with bubble water. I had a similar experience to what I normally take, though seemed to take effect slightly faster than usual and dropped the pain level more quickly (20-25 minutes with reduction occurring over 5 minutes, 6/10 to 2/10, normally 30-40m), and was slightly more sedating possibly because it was already in extracted, aqueous form. I felt a very minor but noteworthy bodily warmth, which I don't normally experience at all--possibly due to 7-OH-MG. Lasted approximately 4 hours, resumed normal use.

It tastes very bitter almost like coffee that has burned with that distinct kratom after taste and put into alcohol (though the preparation was in PG solution), but with less of that "green" plant juice taste, but it was very tolerable. It was syrupy in texture. It came with an eyedropper but it wasn't graduated, so I used a different graduated pipette. It is identical in color to 20x coffee concentrate.

Test 2

Waiting one full day, (normal use), I again stopped all use and achieved a comparable baseline, possibly more lower back pain due to poor sleep position. Resumed all normal medications aside from the muscle relaxant.

I took 1/2 of one Menthol cough drop because I suspected if I had any discomfort in the back of my throat it would be difficult to avoid swallowing the solution based on the amount there was.

Again, I drew 1mL of solution (+/-0.1mL) and put it under my tongue--and my suspicions were correct, it was about the maximum

Sublingual Administration of Sceletium Tortuosum (Kanna) Extract and Parosmia Symptom Relief - My Experience

I recently developed parosmia as a result of an August 2021 COVID-19 infection. I have had symptoms for just short of 3 weeks now.

I occasionally ingest an extract from a South African plant called Sceletium Tortuosum, or Kanna for short, which helps with my undiagnosed anxiety and depression. The last time I ingested this extract was shortly before my COVID infection in August.

I took approximately 70 mg of a 5% alkaloid extract sublingually last night and noticed some mild symptom relief starting about 45 minutes after ingestion. Today, I have virtually no parosmia smell present at all, when it was nearly unbearable just yesterday. I have to really pay attention to get a whiff of the rotten parosmia smell. Food tastes almost normal again, even what were particularly bad flavors like coffee and peanut butter. I would say approximately 90% normalcy if I had to rank flavor against a remembered baseline.

This plant contains a few alkaloids (Mesembrine and Mesembrenone) which act as Phosphodiesterase 4 inhibitors.

I did some digging and found some previous research regarding phosphodiesterase enzyme inhibitors for treating olfactory dysfunction. There is actually a clinical study in the US right now to investigate intranasal theophylline (another PDE inhibitor) for the treatment of post-viral anosmia.

This may entirely be a fluke and I could have started recovering independent of the Kanna but I wanted to get this information out there in hopes that someone with the means might investigate further.

I do not want to encourage people to go take an obscure, unregulated plant/drug so please don't take it based on this post. Please make sure you understand the risks associated with it if you choose to do so. This is more for someone with a medical or pharmacology background to perhaps investigate further.

Hey, just passed a month on Estradiol this week! Got a pedantic question about sublingual administration! :T

A week or two into my hormone treatment, a facebook group convo advised me to dissolve the estradiol under my tongue instead of swallowing it, and my doctor confirmed this may be an idea worth trying out.

Am I losing out on precious, precious estrogen if, while keeping the estradiol pill under my tongue, I check after a few minutes to see if it's dissolved and discover that there's still some grainy goodness under there, which gets licked up and becomes one with my spit?

Is there a better or "official" way to sublingually administer these tasty tasty tit-tacs, to min-max my estrogen levels on blood-testing day in December?

AFAIK s.l. drug administration does not pass the intestines nor any part of the GI-tract. Still my book and wikipedia says it is an enteral (passing the intestines) form of administration. How did we come to this classification and why do we use ist?

Prescribed adderall and taking with / near meals seems to really diminish the effectiveness.

Would it be worth to take sublingual? I know it’s ~20% more bioavailable this route but wondering if that meals tolerance would build up a lot faster also.

First attempt sublingual administration 14 seeds hbwr plus 13 grams fresh magic truffles.

This question is broad, and may lend itself to a product-by-product take, but I'm wondering if anyone has general guidance on which ND powders to take sublingually.

E.g., Gotu Kola seems interesting, but I've seen discussion (e.g., at https://www.reddit.com/r/NootropicsDepot/comments/jx8nxe/gotu_kola_extract_interesting_stuff/) re the rationale for the enteric-coated tablets. First sublingual administration of dextroamphetamine

I need to preface this with my neglect of the chief rule in harm reduction: always start and a low dose. you can always add more but you can never take any away.

15 minutes ago I put two 15mg d-amp tablets under my tongue. within 5 minutes they were disolved and I noticed the first effects but wasn't sure if it was just psychosomatic. 10 minutes later the effects are incredibly pronounced, uncomfortably so. I have a moderate tolerance to amps so I recklessly went ahead and used 2 tablets.

I occasionally will snort a tablet or two as I enjoy the mild rush and the whole process in general, but this is different. I feel weirdly anxious which never happens to me on amps.

ALWAYS START WITH LOW DOSES. You can always take more - you can never take less after you've consumed it.

Evidence for sublingual administration of L-Theanine?

I haven't come across any yet. As far as I know it's just a matter of swallowing it down.

Lately though I've been taking these chewable L-Theanine gummies and holding what I chew underneath my tongue. I know sublingual administration assists with other substances but I'm wondering if the same applies to L-Theanine.

Has anyone here tried administering L-Theanine sublingually. If so, did you notice a difference when compared to just swallowing it right down?

Question about suppository v. sublingual administration.

Since progesterone works best when absorbed through the distal third of the colon, why is E not administered the same as progesterone? If the best absorption happens there why aren’t all medications administered in such a way? Is it because E needs to be dosed multiple times a day to keep levels from crashing? Apparently buccal administration(inside the cheek) makes buprenorphine (Suboxone) x2 more bioavailable(stronger) than sublingual, anyone with experience know how to successfully use it this way?

According to several studies & articles I found, buccal administration of buprenorphine is shown to be two times more bioavailable(stronger) than sublingual administration. I have read peoples personal experiences with this also & they said its certainly stronger. So anyone w/ experience know how to do this most effectively? Do you place it between ur cheek and gums as if it were chewing tobacco or do u put it on ur cheek? I feel like it would touch my teeth or something and fuck it up if I just placed it on my cheek like I read ur β€œsupposed” to do based on pictures I saw but I can’t afford to waste a strip. Sublingual Administration of Ethanol Solution of Estradiol

(I haven't tried this and this is not medical advice. This is a first draft of a concept, not an instruction manual.)

Possible pros of sublingual EtOH solution:
-Don't have to worry about getting it on anyone else, like you would with a topical formula. Good to avoid accidental exposure of someone in your household.
-Greater bioavailability vs. oral administration, since you're mostly bypassing the first pass effect
-Decreased impact on the liver vs oral
-Simple formula with no need for thickeners, gelling agents, or absorption enhancers.
-Fast, easy administration. You can measure a dose with a 1mL dropper or similar
-Fast absorption with no fuss.

Possible cons:
-Potentially irritating if you have a sensitive mouth or are prone to cold sores.
-Causes your blood level of the hormone to shoot up higher, but also drop down faster, than an oral dose. This is okay for some people but not others.
-Not a big improvement over sublingual hormone in pill form.
-You will swallow some of it. This might be an issue if you're on a hormone that's known to be hard on the liver when taken orally.

If this works for any steroid hormone, it should work for all of them. Only use alcohol that you would drink. If you use rubbing alcohol, you'll poison yourself, so don't lol. 40% ABV vodka would work fine. I'd spend a little more for something drinkable that doesn't burn.

The general idea here is that you make a solution of vodka and E2, using serial dilution as necessary to get your desired concentration per mL. Your goal would be to get your dose in 1 or 2 mL. You can measure 1 or 2 mL with a syringe (no needle obviously) or with a feeding syringe that you can get from a pet store.

Sublingual administration is very simple. Stick the dose under the tongue, don't swallow for two minutes, don't eat or drink or rinse for a little while, and that's it. No need for a penetration enhancer, since the mouth has thin skin and is full of blood vessels. It's measurable in your blood within five minutes.

(You could also extract E2 from pills by putting them in vodka overnight to let them digest, and then running the solution through a blender, and then filtering the solution through a coffee filter. This method is most useful if you want to divide your doses down further than you could reasonably split a pill. Say, if you have 1mg pills, but you prefer to take 0.1 mg per day.)

Crushing trazodone for sublingual administration β€” experiences?

Hi, I have dealt with insomnia for around 18 months now after experiencing some major gut dysbiosis. While I've been working on getting my gut back in order, I've been relying on Trazodone to help keep me asleep and achieve REM during the night.

I often wake up after ~6 hours asleep without the ability to fall back asleep. 50mg of Trazodone during these periods help, although, sometimes it's not enough to allow me to fall back asleep. (Trazodone reaches peak plasma after ~1-2 hours after which I'm pretty much awake, albeit tired). I'm wondering if anyone has tried to chew or crush Trazodone for faster administration and whether or not this had been helpful. I may try a smaller dose of 25mg crushed when this happens and see if it works.

Cat taking sublingual Buprenorphine accidentally swallowed it all during administration.

My 5 year old DSH cat had all but 3 canine teeth pulled on Tuesday due to stomatitis, inflammation, and infection. He also has FIV. He is on 5 mg prednisolone per day. As a result, he cannot take an anti inflammatory for pain, such as Onsior. He is currently prescribed 0.15 mL Buprenorphine for pain, to be given sublingually every 12 hours. As I went to administer his nighttime dose, he suddenly jerked his head as I was hitting the plunger, and all of the medicine missed his gums, went into the mouth, and he swallowed it. Other current medications include the Prednisolone every 12 hours and Clindamycin every 12 hours. I have 3 questions.

  1. Will he still experience adequate pain relief?
  2. Is swallowing all the Buprenorphine dangerous?
  3. Should I watch for any issues such as nausea caused by him swallowing the medication? Thanks in advance!

Question about Sublingual HRT administration

So, something of a stupid question, but I recently got prescribed HRT (yay!) and my doctor told me to take it sublingually. This is also good, because I know this is, for many people, a more effective way to take HRT, but now that I've been doing it a few days, I have a stupid question.

Does taking it under the tongue mean sticking the pill to the underside of my tongue? Or do I put it in the cavity directly beneath the tongue? I have not been able to find out and I'm worried I may be doing it wrong.

It's no secret that you can't snort benzos, and sublingual is about as close as you can come. But I'm wondering: how exactly do yall take your benzos?

Personally, I like to crush up a 1mg xan in a piece of paper folded in half, and then pour the resulting powder under my tongue and let it sit for 10 minutes before swallowing.

Question about sublingual administration

I’ve seen posts on here asking whether or not taking (Xanax specifically) is a viable ROA and if it will work..I’ve even done it myself out of curiosity. Most answers I see say that it is a good way to administer and that it will work. The only thing I don’t understand is that since benzos are not water soluble, wouldn’t that mean taking a bar sublingually theoretically shouldn’t work? If the blood vessels in your nose aren’t able to absorb benzos into the blood during insufflation, wouldn’t the same go for the blood vessels under the tongue for sublingual administration? I know there is a certain type of Klonopin tablet that is made to be administered under the tongue, but besides that I don’t understand how putting a bar under your tongue would work other than the excess sludge leftover in your mouth eventually being swallowed and getting into your stomach. Any thoughts or explanation would be greatly appreciated. Hope everyone’s Saturday is going good!

Buccal instead of sublingual administration?

Hi everyone.

I've ordered some Bromantane for the first time, and I've heard that its bioavailability is highest when taken sublingually. However, I've got a fairly small tongue (almost ankyloglossia but not quite diagnosable), which in the past has meant that when I've tried to take things sublingually, I just ended up swallowing them instead.

My question is whether buccal administration (between cheek and gum) works for most sublingual supplements (specifically Bromantane)?

Difficulties with sublingual administration: advice?

My first endo said to take my pills sublingually and my second said it didn't matter, so I went with sublingual.

Here's my problem; when I put it under my tongue, it just doesn't seem to absorb. When I check under my tongue at 10 minutes, I end up with a pool of saliva, and the dissolved pill just sitting in solution. 20 minutes later, I have a truly ludicrous amount of saliva, and while the pool is less blue/green, I think that's because I just produced more saliva and diluted the solution.

Recently, I've tried breaking my morning pill in half and taking a half dose sublingually 6 hours apart, but it didn't help. The estradiol just sits in solution under my tongue. It dissolves, but isn't noticably absorbed.

I try to jam it as far back under my tongue as I can, and seal my tongue with the the sides of the bottom of my mouth so nothing gets out.

Am I doing something wrong? Does anyone have any tips to promote absorption?

Sublingual Estradiol Administration Timing

I'm taking 4mg/day of Estradiol sublingually with no anti-androgens. Since graduating to this dose from 3mg, I have been taking 1mg upon waking, 1mg 5-6 hours later, 1mg 5-6 hours after that, and 1mg directly before bed.

I'm wondering if that's the optimal schedule or if I should take 1mg upon waking, 1mg 8 hours later, and 2mg before bed because testosterone supposedly spikes while sleeping.

Thoughts? I'm trying to hammer down my T the best way possible without AAs.

Powdercities Oxiracetam batch #141211 lot #TW1211. Petrol/Solvent smell, Extremely strange taste, minor chemical burns to mouth, numbness to tongue after sublingual administration.

Being a longterm user and enjoyer of the service and products Powdercity has previously provided, it really and deeply saddens me to have to leave this question, and this warning to anyone else with this same batch of Oxiracetam.

Today I received an order of Oxiracetam in the mail! Excited, I opened package, quickly prepared what I needed. Upon opening the sealed Oxiracetam container I was greeted with a strong, solvent, petrochemical smell.

Because I trust this company, and the fine quality products they've previously provided, I decided "oh well, perhaps its a new synthesis".

Placing my measured 500mg out of the scoop and underneath my tongue, I realized my mistake within minutes. Althought it still had the smallest characteristic sugary taste of oxiracetam, my senses were also assaulted with a an almost immediate chemical foulness, somewhere between the taste or pramiracetam, mixed with PRL-8-53, mixed with phenibut, along with the previously mentioned solvent and petrochemical odour.

It dissolved as it should have in my mouth, quite quickly, while also leaving a persistent numbness to my tongue comparable to dental anesthetic, as well as minor chemical burns to the inside of my mouth.

My numbness has started to fade nearly 4 hours later, while the chemical burn has become more raw, and irritated.

I'm at a complete loss as to what I put in my mouth and what happened. Its as though someone mixed a handful of nootropics into the same tub.

Has anyone else had issues with their Oxiracetam lately? Has anyone else used this batch and lot number??

EDIT/UPDATE 2: Day after. Mouth is still pretty raw, but not nearly as painful as yesterday. Should recover quickly. Using lip chap.

Edit # 3 I've completed a dissolution test on the suspect material after a bit of fear for my safety. 3.5 grams, upon dissolution in water (500ML), produced a CARBONATION type reaction, not giving very visible bubbles, but giving off very audible ones if you place your ears near the reaction vessel.

Pure spring water from a bottle with minimal additives/trace minerals was used to avoid possible contaminants from tap water.

Tmax and sublingual administration of alprazolam

For alprazolam, I saw that Tmax (oral): 1 – 2 hours Tmax (sublingual): 2.8 hours. Why isn't Tmax for sublingual shorter than for oral?

Is oil or alcohol better for sublingual administration?

I'm basically looking to replace bong rips with tincture doses and I want to use distillate to infuse. From my experience, sublingual administration of infused oil takes about 30 min to set it so I was curious if a alcohol solution would work faster and stronger.
