Topiroxostat: a Japanese xanthine-oxidase inhibitor drug similar to allopurinol and febuxostat that can be used to treat gout

I've been having trouble with side effects from both allopurinol and febuxostat, so I did a little research on what other drugs are under development or already on the market. One in particular that caught my eye is topiroxostat (brand names: Topiloric, Uriadec).

From ScienceDirect:

>A novel, nonpurine, selective xanthine-oxidase inhibitor, topiroxostat bears dual inhibitory effects that combine the mechanisms of action of its predecessors. Similar to febuxostat, topiroxostat reduces uric acid production through chemical structure-based inhibition of xanthine oxidase. Akin to allopurinol, the drug covalently binds to, and inhibits, molybdenum-pterin, the active center of xanthine oxidase. Currently approved in Japan at doses ranging from 20 to 80 mg twice daily, topiroxostat 120 mg/day has been shown to be equivalent to allopurinol 200 mg in meeting serum urate target levels.17 Although the half-life of topiroxostat is reported to be 20 h, the agent may have additional, more persistent effects: studies suggest that once inhibited by topiroxostat, xanthine oxidase enzyme function never fully recovers even after disassociation of the drug-enzyme complex.18

It does not appear that this is available in the U.S., and it's not clear to me that it's even widely used in Japan. It caught my eye nonetheless because it's apparently the only xanthine-oxidase inhibitor other than allopurinol and febuxostat that made it onto the market.

I would be very curious to know whether my body tolerates topiroxostat any better than it does allopurinol and febuxostat, but I guess I will never find out since it doesn't seem to be available here.

Does anyone else know anything about this drug, or others that are under development?

Triterpenoid acids from medicinal mushroom Inonotus obliquus (Chaga) alleviate hyperuricemia and inflammation in hyperuricemic mice: Possible inhibitory effects on xanthine oxidase activity , abstract [ 09 - 2021 ]

The purpose of this study was to explore the hypouricemic effect in hyperuricemia mice of triterpenoid acids from Inonotus obliquus (TAIO), and decipher of the underlying xanthine oxidase inhibitory mechanism. Measurement of xanthine oxidase (XO) inhibitory activity was assayed. Organ indexes and serum biochemical indicators were measured in potassium oxonate-induced hyperuricemia mice.

Studies showed that TAIO had the strong inhibitory effect on XO activity, and its inhibition type was mixed and reversible. In vivo, TAIO decreased efficiently uric acid level, hepatic XO, serum blood urea nitrogen activities in hyperuricemia mice. Indicating that TAIO may ameliorate kidney damage and relieve inflammation in hyperuricemic mice, and had the inhibitory effect on XO activity. Furthermore, eight triterpenoids were identified by Ultra performance liquid chromatography electrospray quadrupole time of flight mass spectrometry.

These findings proved that triterpenoids from Inonotus obliquus would have potential biological characteristics and effect on controlling hyperuricemia and gout as an active supplement.

my idea

so i have been wondering about a mix that could give a high similar to weed like lovely nutmeg, and i went to research about it so here is what i came up with black pepper has agonistic activity on TRPV1 receptors which has analgesic properties cause of the release of endocannabinoids like anandamide

black pepper also has a compound called guineensine which is a endocannabinoid reuptake inhibitor and dont forget it has terpenes like B-caryophyllene which bind to the CB receptors

red pepper on the other hand has TRPV1 activity which might mix well with the activity of black pepper

and vanilla which is also a agonist of the TRPV1

we could also add a little Kaempferol to the mix although im unsure from which herb, it has FAAH inhibiting activity

so here is my theory, i truly wonder if it could work and if someone could try i would be glad to hear their experience :) a a black pepper TRPV1a TRPV1 red peppera Kaempferol

i had one article about someone else theorizing about this but i cant find it, i will look harder but its late for me rn and im a bit sleepy i will be glad to talk about it in comments

Sulfur Sensitivity and Candida Connection

Hello everyone! I made a video on the connection between sulfur sensitivity and SIFO (Candida). The idea for this video came from Greg Nigh's book but I might have found a slight mistake in his book. I am no biochemist so I am open to being wrong but he talks about how the enzyme ALDH is molybdenum dependent and breaks down aldehydes and Candida creates aldehydes. I could not find any evidence that ALDH is molybdenum dependent but xanthine oxidase and aldehyde oxidase both break down aldehydes and require molybdenum. So in this video, I talk about how Candida might be holding you back if you have sulfur sensitivity as well as my experience with SIFO and requiring more molybdenum recently, even though my hydrogen sulfide SIBO is in remission

TLDR

Due the complex nature of this company and upcoming catalysts, even this TLDR will be long but I know most of you are not wanting to read 20 pages. Biotech is always complicated and this company has quite a few additional complications, including two uplistings, a reverse split, an IPO and a rapid Phase 3 trial for a treatment for COVID related Acute Kidney Disease.

Their main thesis is that high uric acid causes all sort of metabolic diseases and that lowering it can negate the harmful impacts, there are several studies which support this theory (linked at the bottom).

Xortx has reformulated and improved an FDA approved drug and is repurposing it for the treatment of kidney disease, diabetic nephropathy and acute kidney injury from COVID. They have received very broad patent protection covering ALL treatments which decrease serum uric acid to treat a huge list of metabolic conditions. This patent is much broader than is typically allowed and their greatest asset in my opinion.

Introduction

  • Tea/Lemon Tek could potentiate the effects as more of the prodrug psilocybin converts into the psychoactive psilocin. So more of the psilocin becomes bioavailable.
  • This essentially means you are taking the same dose as you would do by ingesting the normal way but the effects would last over a shorter time-frame.
  • So it is more advisable to start with a lower dose than planned with these methods.
  • More details on how psilocybin metabolises to psilocin: FAQ/Tip 014: Why psilocybin mushrooms/truffles are more sedating than LSD (YMMV)?
  • Also some may experience nausea due to the fibrous chitin which can be hard to digest.
  • This could be partly due to low levels of chitin enzyme, chitinase. More details: 'The Chitin πŸ„ Effect' in FAQ/Tip 002: Have nausea or an upset stomach? Then try 1.5g of ginger. ​
  • Nausea is also a common symptom of the 'come-up' body load effects of macrodosing. So could be an indication that your microdose is too high and you are above your sub-threshold dose^([1]) (or as Paul Stamets more aptly described it as the intoxication dose.)
  • Grinding the mushrooms/truffles into a fine powder increases the surface area which could also potentiate the effects - your stomach or one of the methods below requires less effort to convert the psilocybin, meaning a faster absorption rate.
  • Some may also not like taste of mushrooms/truffles so these methods could help with that.

Cha Qi Feels like Mild Amphetamines

I know Qi from tea is immensely subjective and there are tons of debates over all aspects of it. However, there are some common effects that I see people mention frequently and that I personally experience when drinking Sheng Puer:

  1. increase in body temperature
  2. increase in sweating (I usually become drenched)
  3. palpable increase in heart beat
  4. uplift in mood
  5. stronger mental stimulation than other teas or even coffee
  6. tendency to speak faster and think quicker

I used to take amphetamines for ADHD and these are exactly the same effects experienced from those medications. Coffee usually doesn't have these effects for me unless I have gone a very long time without it and then suddenly drink a lot.

Why would a certain type of tea have similar effects to Amphetamines? What chemicals are in sheng that could contribute to this? Usually caffeine by itself does not cause these effects. I do not get these effects from most other teas, so I doubt it has to do with theanine or polyphenols (the other common psychoactive chemicals in tea). Maybe other xanthines in tea contribute to this effect?

My own theory is that there could be some monoamine oxidase (MAO) inhibitors in sheng that synergize quite well with the caffeine.

I am uninterest in spiritual or metaphysical explanations, although I recognize that these could be meaningful to people.

On my second attack ever. On prednisone and indo which took 90 percent of pain away. Chugging water and cherry juice. Will be looking into allo. I’m on day 12!

SERIOUS: This subreddit needs to understand what a "dad joke" really means.

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.

Dropped my best ever dad joke & no one was around to hear it

For context I'm a Refuse Driver (Garbage man) & today I was on food waste. After I'd tipped I was checking the wagon for any defects when I spotted a lone pea balanced on the lifts.

I said "hey look, an escaPEA"

No one near me but it didn't half make me laugh for a good hour or so!

Have sulfur sensitivity? Candida might be holding you back

I recently started needing more and more molybdenum and I was backsliding and regressing. I thought my hydrogen sulfide SIBO must be back but when I realized it wasn't, I went looking for more answers for why I might need gobs of molybdenum so I made a video on the connection between sulfur sensitivity and SIFO (Candida). The idea for this video came from Greg Nigh's book but I might have found a slight mistake in his book. I am no biochemist so I am open to being wrong but he talks about how the enzyme ALDH is molybdenum dependent and breaks down aldehydes and Candida creates aldehydes. I could not find any evidence that ALDH is molybdenum dependent but xanthine oxidase and aldehyde oxidase both break down aldehydes and require molybdenum. So in this video, I talk about how Candida might be holding you back if you have sulfur sensitivity as well as my experience with SIFO and requiring more molybdenum recently, even though my hydrogen sulfide SIBO is in remission

Why should I participate?

For the opportunity to WIN this month's object of the month! Winners will receive the OOTM Award (equivalent to a platinum award), a special winners flair, and your image can be added to the banner rotation at the top of the subreddit! You will also be able to choose the image of the month for the next month from a shortlist. (This is to ensure that it as a target that will be up at night this particular month, as well as ensure that there is a good mix between small and large objects).

How can I participate?

Firstly just submit your image to the Subreddit like normal. If you want to participate in the contest, leave a comment in this thread with the link to your post. On the last 3 days of the month, this thread will be replaced by a voting thread where anyone can vote on what image they think should win. When the voting thread is up no new posts will be allowed to enter into the contest. This allows for all posts submitted to receive an equal amount of time in the voting thread. Note: ONLY IMAGES CAPTURED IN THE LAST 6 MONTHS WILL BE ALLOWED TO ENTER. You can make multiple entries, as long as you are submitting images from different datasets.
