Introduction of α‐N‐methylated non‐proteinogenic amino acids into peptides can improve their biological activities, membrane permeability and proteolytic stability. This is commonly achieved, in nature and in the lab, by assembling pre‐methylated amino acids. The more appealing route of methylating amide bonds is challenging. Biology has evolved an α‐N‐automethylating enzyme, OphMA, which acts on the amide bonds of peptides fused to its C‐terminus. Due to the ribosomal biosynthesis of its substrate, the activity of this enzyme towards peptides with non‐proteinogenic amino acids has not been addressed. An engineered OphMA, intein‐mediated protein ligation and solid‐phase peptide synthesis have allowed us to demonstrate the methylation of amide bonds in the context of non‐natural amides. This approach may have application in the biotechnological production of therapeutic peptides.

I have been taking high doses of glycine (30g) every day for several years. I always assumed it was safe since it's an amino acid found in high levels in foods. Any thoughts?

I have a sleep disorder where I do not enter deep sleep properly (my sleep study showed I dipped moved from light sleep to REM with little stage 3 or 4). After years of research I discovered glycine promotes these stages of sleep that I was missing. Now I have almost completely managed to control it with high doses of glycine (15g, twice nightly, 30g total). I buy AjiPure since it's the only pharmaceutical grade glycine I can find. I assumed it was safe since glycine is commonly found in high amounts in foods. Any reason to think I might be wrong?

If I have trouble remembering the V I just think β€œvagina”. WAP stands for the cardi B song! If you want to include glycine you could also do:

Non-essential amino acids: A possibly misleading misnomer

For an amino acid to be considered non-essential it needs to not be produced in 'significant' quantities within the human body. This is what keeps some essential amino acids from being considered non-essential, since some are produced in very small quantities. However, the criteria for 'significant' is unestablished. It is possible that some amino acids may be misleadingly classified as 'non-essential' because they are produced in the human body, but not in optimal quantities.

It may be beneficial to intake certain non-essential amino acids to supplement their inadequate endogenous production, but I am unable to find research on this aside from this paper (which talks more generally about mammals).

Any research/speculation anyone could offer on this topic would be much appreciated.

Warm cozy feeling 30-60 minutes after taking large doses of amino acid supplements

Whenever I take a large dose of any of the follow supplements I get this warm/hot cozy feeling inside.

Gaba, agmutine sulphate,NAC, taurine

It feels like it is coming from the inside of me though, like I have no desire to take off my jacket or regulate my external temperature. It feels really nice and relaxing and lasts for around 20 minutes. It makes my breathing kinda weird idrk how to describe it, but if it happens during a workout then I just sit down for a few minutes.

Anyone know what this is?

[Routine Help] dehydrated skin: hemi squalene oil or amino acids + b5, or both?

hello!

i have very dry, sensitive skin (eczema-prone on and around the eyelids) and i'm absolutely terrible at keeping up skincare routines (for this reason i'm contemplating buying smaller sizes and reducing waste). i'm also on a budget, and i gravitate towards greener options in terms of production and packaging

i don't have a skincare routine right now, but i decided to try the ordinary's squalene cleanser and their moisturiser. i've used their hyaluronic acid in the past, and while it was good, i'm curious to see if their plant-derived hemi squalene or their amino acids + b5 are better alternatives – i've seen equal amounts of good reviews for both, so i don't really know where to go from here

Justifying a chemistry class I once took by adjusting Mordin's Amino Acid song...

Though the song is excellent, there is an unfortunate error that I think we can update. Here are the original lyrics:

>Glycine, valine, leucine, isoleucine, alanine, they all are aliphatic, so you will not see a ring!

>The lone human amino acid with one is proline. From protein we are formed!

>With two hydroxyl groups you'll find serine and threonine. For sulfur, look to cysteine or to methionine!

>Tryptophan and tyrosine and phenylalanine! Aspartic and glutamic are both acids as we've seen.
>
>For amides, see asparagine and also glutamine. From protein we are formed!

The problem:

"They all are aliphatic, so you will not see a ring" This statement is technically true; those 5 amino acids are aliphatic and do not contain any ring structures (heteroatoms such as carbons linked together in an enclosed ring). The issue is this implies that Aliphatic (non-aromatic) molecules NEVER have rings and ONLY Aromatic molecules have them. This is completely wrong, especially in light of the next line. "The lone human amino acid with one is Proline" which is wrong because Proline is NOT the only amino acid with a ring structure (All the aromatic AAs have ring structures), and it is NOT Aromatic. So if we were to update the line to be chemically accurate it could go something like:

>Glycine, valine, leucine, isoleucine, alanine, they all are aliphatic, AND you will not see a ring!
>
>The only aliphatic acid WITH a ring's proline. From protein we are formed!

That way the statement about proline is accurate and we aren't erroneously implying that aliphatics never have rings or that proline has the only ring out of all the amino acids.

Anybody gotten an amino acid level/deficiency panel run? My new theory

Alright so I've had a lot of success treating many of my symptoms by supplementing with amino acids and anti inflammatories. Since I still have some symptoms left I was checking out a list of amino acids and their function to see if anything else would be worth trying. Honestly found a number that I thought seemed good. I've also read a lot about how amino acids can be depleted during times of stress and illness, and it can be hard to get individual amino acids back to a normal balance without direct supplementation of that amino acid.

So anyway I'm kind of formulating a new theory that atleast for my longhaul symptoms I either have lingering inflammation from covid stressing my body out causing protein/amino acid breakdown issues and imbalances or I have amino acid imbalances causing stress causing inflammation. Vicious cycle

Has anybody had any kind of amino acid levels testing done? They're definitely not routine at the doctor and kind of pricy, also not sure if they're super accurate; but I felt like if there was a glaring issue we saw in a lot of people that could shed some light on what's going on with longhaulers; maybe even break the cycle of stress and inflammation by addressing the imbalances.

Also I have no medical knowledge whatsoever this theory just came to me trying to research new things I could try to help my symptoms. Felt like somewhere I've heard ~every disease is caused by a nutritional deficiency/imbalance~ which got my gears turning.

Request: is there a reliable list of dietary amino acids interaction?

For example we know that tryptophan competes with other large neutral amino acids for the BBB but what about other amino acids?

For example if I take a tablet of X-amino acid + 1 of Y-amino acid, will they compete or act synergistically? I'm looking for a reliable table of all amino acid interactions, if it does exist.

Some amino acids can be used as a supplement, for example L-glutamine. Would it harm/change the molecule when adding it to hot beverages, like tea?
