(Nasdaq: SNGX) (Soligenix or the Company), a late-stage biopharmaceutical company focused on developing and commercializing products to treat rare diseases where there is an unmet medical need, announced today that following the validation of synthetic hypericin's biologic activity in the positive pivotal Phase 3 FLASH (Fluorescent Light Activated Synthetic Hypericin) study in cutaneous T-cell lymphoma (CTCL), as well as positive proof-of-concept (PoC) demonstrated in a small Phase 1/2 pilot study in mild-to-moderate psoriasis patients, the Company will be expanding this novel therapy under the research name SGX302 into psoriasis, a large and underserved market with a significant unmet medical need.

Visible light-activated synthetic hypericin is a novel, first-in-class, photodynamic therapy (PDT) that is expected to avoid much of the long-term risks associated with other PDT treatments. Synthetic hypericin is a potent photosensitizer that is topically applied to skin lesions and taken up by cutaneous T-cells. With subsequent activation by safe, visible light, T-cell apoptosis is induced, addressing the root cause of psoriasis lesions. Other PDTs have shown efficacy in psoriasis with a similar apoptotic mechanism, albeit using ultraviolet (UV) light associated with more severe potential long-term toxicities. The use of visible light in the red-yellow spectrum has the advantage of deeper penetration into the skin (much more than UV light) potentially treating deeper skin disease and thicker plaques and lesions, similar to what was observed in the positive Phase 3 FLASH study in CTCL. Synthetic hypericin or HyBryte(TM) (tradename used in CTCL) was demonstrated in this study to be equally effective in treating both plaque (42% treatment response rate after 12 weeks treatment, p<0.0001 relative to placebo treatment) and patch (37%, p=0.0009) lesions of this orphan disease caused by malignant T-cells. Further, this treatment approach avoids the risk of secondary malignancies (including melanoma) inherent with both the frequently used DNA-damaging drugs and other phototherapies that are dependent on UV A or B exposure. The use of synthetic hypericin coupled with safe, visible light also avoids the risk of serious infections and cancer associated with the systemic immunosuppressive treatments used in psoriasis.

Hello!

I was wondering If you guys had any knowledge of the safety for women Using birth control to take St Johns wort without the active ingredient (hypericin/hyperforin) in it?

Ive gotten a little concerned as Im on Hormonal birth control (nexplanon) And was told that st Johns wort can be found in hypericin-free alcohol distillate? I was wondering What that is And where that could be found?

Hypericin or Hyperforin?

Hi, I hope this is the correct subreddit for this question.

Due to some sleep problems and anxiety I have been having on & off since November, I'm thinking of trying St. John's Wort. I don't want to use a prescription medicine until there are no other options left. My question is, is hypericin proved to be completely inefficient for treating anxiety & depression? I have read lots of scientific papers where it's said that hypericin was thought to be the major agent of St. John's Wort's antidepressant effects, but it turned out that it was actually hyperforin. The problem is, there are no hyperforin-based products in my country. I can order them from Amazon but it would take at least two weeks and cost a lot of money. If I buy a hypericin-based, standardized product, would it help me combat my anxiety and sleep problems at all?

FYI, my main problem is waking up after just 5 hours of sleep every night. I can go to sleep very easily, but no matter what time I go to bed, I always wake up after maximum 6 hours (and 6 is very rare, it's almost always 5 hours). When I wake up, my whole body - especially my legs- are tense, and most of the time I feel very nervous. Getting back to sleep is very rarely possible, and I just lay awake until the alarm goes off. I believe my anxiety stems from this sleep deprivation, because I have a lot difficulty concentrating and it makes me feel nervous (and sometimes even panicky) at work, thinking that I will make a big mistake or will let all my colleagues down. I also tried 5-HTP; it seemed to work great for a week, but then it made me feel worse, so I stopped it.

TL;DR: Is hypericin proved to be completely useless? Is it really hyperforin that does all the work in St. John's Wort?

Supercritical CO2 Saint John Wort extract a.k.a. Hyperforin without hypericin

I would be very interested if ceretropic would produce preparations including super CO2 Saint John wort extract WITHOUT hypericin

Indeed, this method of extraction allows filtering out hypericin, which was thought for a long time to be the major active compound of SJW, but has since then be retouted to be only a part of its anti depressant effects.

Since there are concerns about the photosensitivity induced by hypericin and doubts about it favorising cataracts (not necessarily backed-up by strong and reliable research though, but I don't know about you but it'd rather take the safest option when it involves the safety of my sight)

I think it is time some company offers the market a product focused on hyperforin and hypericin-free

From what I've seen over the Internet, CO2 supercritical extracts are only sold to businesses or for wholesale, and pure hyperforin from labs is expensive (I don't know if it's because it is synthetised or if removing the other CO2-soluble compounds makes it costly).

Considering SJW is one of the most popular, if not the most self-medicated antidepressant out there, I think it's time to give an option to people to get a strong hyperforin dosage without the negatives effects of hypericin

I found a 180 pages long phD work reviewing studies on SJW and it says that over 2 g of hypericin a day, photosensitivity starts to be an issue - thus, it is limiting the max amount of hyperforin one could take without worrying/experiencing issues

What do you guys think ?

SERIOUS: This subreddit needs to understand what a "dad joke" really means.

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.

Again, to all the fellow dads, I apologise if I'm sounding too harsh. But I just needed to get it off my chest.

Ive been taking perika, which is a formulation of st johns wort, for my anxiety for a while, and Ive been trying to find other vendors that sell it that change their formulations to better suit hyperforin, which is what is said to target anxiety and doesnt cause the skin issues seen in hypericin. Do you guys sell that? do you guys plan to bring it in to your online store?

Based on the biological proof of principle shown both nonclinically and clinically with dusquetide, a novel synthetic peptide that modulates the body's innate immune system, Soligenix continues to explore product opportunities, both in the reduction of oral mucositis in HNC and as a potential anti-cancer treatment. Dusquetide binds to p62 or SQSTM-1, a scaffold protein implicated in a number of intracellular signaling networks implicated in tumor cell survival, including autophagy. The role of p62 is best characterized in multiple myeloma and breast cancer. All variants of breast cancer, including metastatic breast cancer, estrogen receptor positive (ER+), human epidermal growth factor receptor 2 high expressing (HER2+) and triple negative expressing cell lines, have demonstrated a significant role for p62 in tumorigenesis.

The MCF-7 cell line tested in the xenograft studies with dusquetide is both ER+ and responsive to anti-HER2 treatment. Treatment with dusquetide was effective not only as a stand-alone treatment (p<0.01 for tumor size), but also in conjunction with radiation (p<0.05 vs radiation only for survival), chemotherapy (paclitaxel) and targeted treatment (trastuzumab; p<0.001 vs. placebo only for tumor size), reducing tumor size and enhancing overall survival. Other tumor types also have been shown to be dependent on p62 express
