Potassium Bromide: obscure cough syrup trip report

The first time I heard about potassium bromide was only five days ago. I tried searching for the mentions of it and found out that KBr is still used in my country as one of the active ingredients in a cough syrup called Pertussin, which contains 12g thyme extract and 1g KBr and comes in 100ml bottle. It is fairly rare here, I had to place my order online with local pharmacy in-store pickup. Supposedly it was used quite frequently in the Soviet Union but eventually was replaced by safer drugs. Even though bromides were succeeded by newer drugs and prohibited for use as medication in most countries, they weren't banned in USSR and Russia as they were never used as sedative or anticonvulsant meds, making bromism cases non-existent. There's no information on recreational use of Pertussin in Russia as it was always overshadowed by other OTC meds, especially barbiturates like Corvalol

link to a Russian pharm wiki, use google translate: vidal.ru/drugs/pertussin__30476

4 bottles cost me 200 rubles (2,60$) but you can buy it as cheap as 0.20$ for one bottle. This syrup kinda tastes like Pepsi. I poured some in my cup and mixed up with Sprite. My buddy took 1g and I had to take 2g since one of my friends started feeling nauseous. He's probably allergic to thyme. I was sipping slow and it started kicking in 15 minutes later. Feels like low dose benzo with sleep deprivation euphoria but you remain functional, it def calms your mind. We were on top of nine-story building watching sun set and talking and even managed to climb down easily. After coming home I decided to listen to some music. Felt good but nothing special, you just feel relaxed and all. Then I went to bed and passed out in a minute. Had great sleep, woke up still feeling calm with no anxiety at all

So that's it. I would like to try it again but with higher dosage, like 3 or 4 grams. My friend who took 1g only felt relaxed and said effects were underwhelming, so I recommend taking 3 grams for the first time.

I forgot to keep track of time and probably made some spelling and punctuation mistakes in this text. The next non-rx drug I'm looking to try is Andipal, which contains 20 mg phenobarbital and 250 mg of metamizole aka dipyrone.

Bromide Salts?

There seems to be a consensus that bromide salts can cause some type of CNS depression and euphoria. Both potassium bromide and sodium bromide were used medically for their hypnotic, anticonvulsant and sedative effects throughout the late nineteenth century and into the twentieth century where they were promptly succeeded by barbiturates and eventually benzodiazepines. Obviously the regular intake of bromine is terrible for you and these medications had severe side-effects when used daily due to a lengthy half-life of ~12 days where bromine would accumulate in the body and eventually people would present with bromism.

Despite all this there is a sparse wikipedia page, nothing on Erowid or PN Wiki and a measly 3 or 4 reddit threads, of which only one is a trip report, on these drugs. Presumably, using very sparsely and not taking massive doses, they could be used safely. Has anyone used or know anything about the bromide salts?

Difficulties with N-Alkylations using alkyl bromides

I've had to run a few reactions of this type over the past while, and they've been consistently giving me some difficulties. Specifically, I've found that they don't tend to proceed to completion, leaving a lot of unreacted starting material, based on crude LC-MS. I think this is also contributing to purification difficulties. The literature I've been working from seems to claim very high yields (>90 %), so I'm wondering if I'm missing something.

The first example is this reaction. The first procedure gave yields below 25%, despite the reference claiming above 90% (https://doi.org/10.1002/ejoc.200701190). My scale was 10-fold lower than theirs, but I'm not sure how that would affect the yield. I also tried using ACN as a solvent instead, but this barely gave any trace of product. I eventually landed on the second procedure (https://doi.org/10.1039/C5CC05395A, in Supporting Info), through which i was able to get a 45% yield, roughly half of what they claim. Purification was a bit of a nightmare, as the cooled reaction formed a rock hard mass that I had to use a bunch of methanol to get out of the flask. I was eventually able to get the product to crash out using acetone of all things, and the yield was enough to move forward, despite being underwhelming.

Recently, I've been working on another reaction that also involves N- (and O-) alkylation using an alkyl bromide, and also seems to not work. The reference (https://doi.org/10.1016/j.bioorg.2017.03.002) doesn't explicitly state the yield of this step, but they strongly imply that it was high, based on the quantities used in the next step. What I found was that neither the starting material or the K2CO3 were particularly soluble in the solvent (acetone), and that, at most, trace amounts of product were generated, with huge amounts of leftover starting material. As a potential solution, I've considered changing to a better solvent (maybe ACN?) or using a more soluble base (cesium carbonate). I also have access to a microwave reactor, which has helped with some reactions in the past.

I know the mechanisms of the reactions are likely quite different, one using base and the other not, but I'm wondering whether there's a similar issue causing me problems. I don't think the issue lies with my lab practices (I've used dry solvents, reagents, and glassware), but I'm curious whether there's any reason these reactions should be giving me trouble, or if th

Sodium Bromide Trip Report

(TL;DR at bottom) So I was recently reminded to try this substance by another post about bromide salts and their past usage as a sedative hypnotic. I bought some NaBr a few months ago with the intent on trying it but honestly forgot I owned it until recently. Before you read I feel lile saying this report is a little boring as the effects were pretty subtle and underhwhelming.

8:35 measured 2.2grams of NaBr and parachuted. 8:53 feels like it might be starting to kick in. Slight change in perception that's hard to put a finger on although this may just be placebo. Also feeling a mild floaty feeling in my head. I feel more physically sedated as well- my body feels relaxed but I don't feel sleepy. 9:54 Just finished eating dinner and perceptual change seems to have faded or was likely just placebo. The effects so far have been very subtle with only some mild sedation. 10:58 No longer noticing any effects although they were not very pronounced to begin with. I would group this with other compounds like chlorobutanol or even chamomile tea which have subtle effects and could perhaps be helpful if trying to sleep or relax.

Well there you go, it didnt make me super drowsy or feel like I could go right to sleep like from taking melatonin or from the times I've tried chlorobutanol (another old hypnotic sedative). I don't think I'll be trying NaBr again but hopefully this experience can give some insight on this rarely documented compound.

TLDR: 2.2 grams of sodium bromide (old hypnotic sedative) caused only mild sedation and was underwhelming.

DXM, Bromide, and the Thyroid - Re-Evaluating the Risks of HBr

Note: You may notice my account was created just today. This is because I don't want my main reddit account associated with recreational DXM use. Hope you understand :)

I wanted to post this as a warning for people who use DXM regularly. While "week per plat" is talked about, I'm certain for every 10 people that follow that rule, there is at least 1 person who doesn't. That being said, I was taking DXM HBr about 2-3 times per week in high 1st to low 2nd plateau doses (roughly 240-480 mg on average) as well as taking a prescribed medication which contains an HBr salt. I'll probably get scolded for this, but it's made a huge difference in my mental and spiritual health, has made me less stressed and hateful, and I no longer have to take a cocktail of antidepressants which never worked well in the first place. However, that isn't what I'm here to talk about, and I don't want relevant background info to be conflated with me glorifying poor harm reduction practices, but quite the contrary: I'm here to talk about the possible negative health effects of Bromide intake.

A friend of mine calculated that, between DXM and my medication which do not interact otherwise besides both containing Bromide salts I was consuming on average 700 mg bromide salts per week, or ~100 mg per day in a worst case scenario. If you adjust this number for diet, it equates to over 450 mg DXM Hbr per day. The half life of bromide in the body is 9-12 days, with some source claiming it's even as high as 14 d, and even 25d on a low sodium diet! Using the half life formula, after 3 months, I would have over a gram of bromide in my blood! The average person intakes up to 8mg bromide per day from the diet. By comparison, that's over 10 times more bromide! Maybe not enough to cause bromism, but it certainly can't be good for you.

The full text for this article (pubmed.ncbi.nlm.nih.gov/12948087/), which my friend was able to get, is a case report of a much more intense case of DXM abuse with another Bromide containing drug, but that's not really relevant. What is relevant that it cites another article (which is in a book and he couldn't find it online) which states that relative values of bromide toxicity can begin at as low as 50-150 mg/dL. Considering I don't eat very much sodium at all, just hypothetically, using the highest half life for bromide of 25d sticks me at 75mg/dL, and this is just for toxic effects. Using the lowest half life of

How much (if any) do you cool your agarose sltn before adding ethidium bromide?

Hey all, I've been troubleshooting my shite PCR images.

In a previous lab we added a different fluorophore (I don't remember what it was), and we did not cool the gel before adding the dye. Whereas now in my current lab we run the flask used to heat the mixture under cool water, cooling the mixture before adding EtBr. I've been told this prevents the decomposition of the EtBr.

Is this necessary? I'm worried that I'm cooling it too much, or not consistently, in part resulting in my variably shite to good images. Any thoughts?

Has anyone been prescribed this? I have been using Flonase to reduce my polyps, but my doctor said my polyps are like grapes. She prescribed the above to shrink my polyps, has anyone here used it before? Just looking for others experience using it.

UPDATE: Well, it has been 13 days and so far I feel as though it has helped a bit. I would like to have the doctor actually look and see if my polyps have shrunk or not. I don't know if I am fooling myself or not, because I do feel as though I can breathe through my nose better. I feel as though I am doing a lot less mouth breathing due to blocked nasal passages. Although, according to my wife it hasn't helped all that significantly with my snoring, I think it has helped shrink the polyps significantly enough to give me a bit of relief. Whether it's short or longer term, we'll see.

Scalable Negishi Coupling between Organozinc Compounds and (Hetero)Aryl Bromides under Aerobic Conditions when using Bulk Water or Deep Eutectic Solvents with no Additional Ligands

Coupling runs across water and DES: Scalable Pd‐catalyzed Negishi cross‐couplings between (hetero)aryl bromides and organozinc halides have been developed either by using bulk water, or the choline chloride/urea eutectic mixture as a reaction medium. These processes proceed under mild conditions, short reaction times in air, in the absence of additional ligands, with high chemoselectivity and a broad substrate scope.

Abstract

Pd‐catalyzed Negishi cross‐coupling reactions between organozinc compounds and (hetero)aryl bromides have been reported when using bulk water as the reaction medium in the presence of NaCl or the biodegradable choline chloride/urea eutectic mixture. Both C(sp3)‐C(sp2) and C(sp2)‐C(sp2) couplings have been found to proceed smoothly, with high chemoselectivity, under mild conditions (room temperature or 60 °C) in air, and in competition with protonolysis. Additional benefits include very short reaction times (20β€…s), good to excellent yields (up to 98 %), wide substrate scope, and the tolerance of a variety of functional groups. The proposed novel protocol is scalable, and the practicability of the method is further highlighted by an easy recycling of both the catalyst and the eutectic mixture or water.
