RNA-Seq ? : isoforms via post-processing of mRNA?

Does RNA-Seq protocols deal with pre and Mature RNA after all the eukaryotic post-processing? or is it mixed and dealt with afterwards with QC scripts?

What is the difference between "major" and "minor" isoform of a protein?

I'm looking at two sequences for HSP70 for the same species and from the same authors in ncbi. The only difference in the two titles are the terms "major" vs. "minor form". What exactly defines the protein as major as opposed to minor form? Is the major form more commonly expressed than the minor one?

SMRT vs ONT: long read transcriptomics for novel isoform dicovery and differential transcript usage.

Hello everyone,

I am a bioinformatician who is relatively new to long reads sequencing technologies, and I was interested in its application to transcriptomics. I would be very interested in discovering the differences in isoforms proportion between, for instance, control vs. patient.

I did a fair amount of research and was pleased that there are many bioinformatics tools available.

I am, however, not sure about which sequencing technology is best suited for that kind of application.

As far as I understood, on the one hand, we got SMRT from PacBio producing quality long reads with more or less 10kb length. And on the other hand, we got Oxford Nanopore which can produce very long reads up to 1 million bases but with a higher error rate.

Can anyone who had experience in transcriptomic with one or both technology give feedback on the pros and cons, or difficulties, you have experienced?
