Running 95% formamide on agarose gel

Hello!

Does anyone have any experience running high percentage formamide solutions directly on agarose gels?

I am eluting biotinylated DNA from Dynabeads MyOne Streptavidin T1, which requires elution in 95% formamide + 10 mM EDTA (According to Invitrogen). My question is rather or not I can run these samples directly on the gel or if I need to do an EtOH precipitation first. I have seen examples and protocols of people running 60% formamide solutions, but nothing higher than that.

Any reason why formamide might degrade into something nasty under electric current?

I am a biochemist/structural biologist graduate student and I'm currently working on an RNA project that is giving me some problems in regards to RNA degredation. I believe this degredation is due to contamination with RNAses. I know that formamide can protect RNA against RNAse degredation and I typically use formamide at a 50% concentration in all of my buffers to help with that. One step, however, involves extraction of RNA from a slice of acrylamide gel by placing the slice in an electric field and "electro-eluting" the RNA out of the slice into a dialysis bag of buffer. This buffer does not include formamide and is the most likely place where RNAses are degrading my RNA. I was wondering, if I included formamide in the electro-elution, if there would be any risk that the formamide might decompose into something nasty that would either be bad for me or for my RNA. The composition of the buffer is 20 mM Tris pH 7.5, 10 mM acetic acid, 500 uM EDTA. Is there any reason to believe under 100 V (couldn't tell you the number of amps) the formamide would degrade?

Hello women of science

I am a developmental biologist... and now also an experiential embryologist! i.e. I am going to have a baby. I do a lot of in situ hybridizations, which use formamide, which is a teratogen (boo!)

Does anyone know of in situ protocols that do not require formamide?

SERIOUS: This subreddit needs to understand what a "dad joke" really means.

I don't want to step on anybody's toes here, but the amount of non-dad jokes here in this subreddit really annoys me. First of all, dad jokes CAN be NSFW, it clearly says so in the sub rules. Secondly, it doesn't automatically make it a dad joke if it's from a conversation between you and your child. Most importantly, the jokes that your CHILDREN tell YOU are not dad jokes. The point of a dad joke is that it's so cheesy only a dad who's trying to be funny would make such a joke. That's it. They are stupid plays on words, lame puns and so on. There has to be a clever pun or wordplay for it to be considered a dad joke.

Again, to all the fellow dads, I apologise if I'm sounding too harsh. But I just needed to get it off my chest.
