Beta catenin luciferase reporters

I’ve been trying to use the top flash reporter assay. Seems to work but my renilla (PRLTK) always goes up alongside the firefly signal, I’m talking a huge increase. Are there other renilla reporters I could use or a dual firefly/renilla construct?

Ratio of firefly to renilla is 20:1 I’ve also tried 40:1 same issue.

Steric hinderance by active/ total beta-catenin antibodies on same IR western blot?

I have been using the Li-Cor IR western system to test ratios of active to total beta-catenin, using both antibodies (from different animals) and different color secondaries on the same blot. I was wondering if it is possible that the two antibodies are large enough to hinder each other from binding, thus altering my results?

Does Wnt/beta-catenin signalling in stem cells, be it embryonic or adult, promotes differentiation or maintain stem cell self-renewal?

I am confused by this Wnt signalling concept. On one hand I read about Wnt signalling is required for stem cells to maintain their stemness. On the other hand, I've read that Wnt promotes follicular stem cells to drive hair formation.

If it happens to be both cases could you provide a specific example where the signalling pathway promotes differentiation or maintain stem cell self-renewal?

So when I played the BETA, people told me "it's a 3 month old build" and "i should wait for a day 1 patch" .... If you said this, how are you enjoying the game?

Inquiring minds would like to know. I said "even if this is a 3 month old build, the game is in a rough state and there is NO WAY that issues could be fixed in 3 months".

But y'all kept going on and on and on about how its a 3 month old build

So, if you defended the BETA, how are you enjoying your fully fleshed out Battlefield? :) Are you "having a blast?"
